What time of year are you doing this? That may affect my advice a bit. But assuming this is not deep summer, I think you have a great plan. Here's how I would do the trip from Sanibel to Key Largo -- Drive on US 41 Tamiami Trail, which takes you through the heart of the Everglades. You can take an airboat ride along here. When you get to Florida City (towards the end of the drive), go to Everglades National Park and walk the Anhinga Trail (1/2 mile boardwalk). Lots of birds and gators.
And absolutely include Miami Beach in this trip.

The original Universal Studios Florida and the Islands of Adventure are the two main theme parks of this resort.
